Abstract:Objective To investigate the expression pattern of sialic acid-binding immunoglobulin-like lectin 15 (Siglec- 15) in laryngeal squamous cell carcinoma (LSCC) and its correlation with clinical characteristics. Methods The UCSCXena data base was accessed to obtain the gene expression profiles and clinical data of LSCC from The Cancer Genome Atlas (TCGA) database. Additionally, tumor tissues and adjacent tissues from 49 LSCC patients admitted to the Affiliated Hospital of Nantong University between 2000 and 2009 were collected for immunohistochemistry (IHC) detection. Correlation analysis was performed between Siglec-15 expression and key clinical indicators. Results Bioinformatics analysis revealed that Siglec-15 expression was significantly upregulated in LSCC compared with adjacent tissues. Clinical data from TCGA showed that the overall survival of patients with high Siglec-15 expression was significantly shorter than that of patients with low expression [368 (230, 589) days vs 495 (286, 1 472) days, P=0.034]. Kaplan- Meier survival analysis performed on 49 patients with LSCC indicated that high Siglec- 15 expression (P=0.021), female gender (P=0.012), and positive lymph node metastasis (P=0.002) were associated with poorer prognosis. Conclusion Siglec-15 is abnormally highly expressed in LSCC and is correlated with positive lymph node metastasis and poor survival outcomes.
