中文摘要: 目的 运用16S rDNA高通量测序技术,分析二尖瓣脱垂(MVP)患者肠道菌群结构组成;与健康人群进行对比,进行物种多样性、差异性分析,探索差异菌属与MVP病变的相关性。方法 选择2023年1月至2024年6月江苏省人民医院因MVP拟行手术治疗的患者[(MVP组,n=10)及招募年龄、性别匹配的健康人群(对照组,n=10)]作为研究对象,采用16S rDNA高通量测序技术检测两组的肠道微生物群落结构。结果 MVP组肠道内的操作分类单元(OTU)数目高于对照组;两组肠道菌群的α多样性差异无统计学意义(P>0.05);两组肠道菌群的β多样性差异具有统计学意义(P<0.05);MVP患者的芽孢杆菌纲、β?变形菌目、乳杆菌目、氨基酸球菌科、拟杆菌科和拟杆菌属的丰度高于健康人群肠道菌群,差异有统计学意义(P<0.05)。LEfse分析显示,MVP组肠道菌群中巴斯德菌目、巴斯德杆菌科、嗜血杆菌属、毛螺菌属的丰度在菌群差异中起主要作用。结论 MVP者与健康人群的肠道菌群丰富度存在差异,但是整体群落多样性并无显著差异,MVP患者存在的特定的菌群可能与MVP的发生发展相关。
Abstract:Objective To investigate the gut microbiota composition in patients with mitral valve prolapse(MVP)using16S rDNA high?throughput sequencing technology,and to analyze species diversity and differential taxa between MVP patients and healthy controls, thereby assessing the correlation between altered microbial genera and MVP pathogenesis. Methods A total of 20 participants were recruited,including 10 MVP patients(MVP group)scheduled for surgical treatment at Jiangsu Provincial People's Hospital from January 2023 to June 2024,and 10 age? and gender?matched healthy controls (CON group). Gut microbiota profiles were analyzed using 16S rDNA high- through put sequencing technology. Results The MVP group exhibited a higher number of operational taxonomic units(OTUs)compared to controls. No statistically significant differences were observed in alpha diversity(P>0.05). Significant differences in beta diversity between groups were detected(P<0.05). The abundances of Bacilli,Betaproteobacteriales,Lactobacillales,Acid aminococcaceae,Bacteroidaceae,and Bacteroides in the gut microbiota of patients with mitral valveprolapse were significantly higher than those in healthy individuals(P<0.05). LEfse analysis revealed that the abundances of Pasteurellales,Pasteurellaceae,Haemophilus,and Lachnospiraceae_UCG?001 in the gut microbiota of the MVP groupplayed a dominant role in the microbiota differences. Conclusion There is a difference in the gut microbiota richness between patients with MVP and healthy individuals,but no significant difference in the overall community diversity.The specific microbiota present in patients with MVP may be associated with the occurrence and development of the condition.
