中文摘要: 遗传性心律失常(IA)是由离子通道及相关结构/钙调控蛋白基因异常引起,其典型特征包括家族聚集性和高猝死风险,严重干扰心脏电活动的产生与传导。微小RNA(miRNA)作为一种内源性短链非编码RNA,在转录后负向调控基因表达,对维持心脏电生理稳态和结构完整性具有关键作用。miRNA 表达失调与IA的发病机制密切相关,一旦miRNA 功能受到干扰,将直接影响心脏的节律和收缩功能,进而诱发心律失常。因此,深入探究心脏特异性miRNA的表达模式和调控机制,对于揭示IA的病理机制及开发新型治疗策略具有重大意义。
Abstract:Inherited arrhythmias(IA)are a group of genetic disorders involving ion channels,calcium handling proteins,and cardiomyopathic substrates,which is typically characterized by familial aggregation and a high risk of sudden death,severely disrupting the generation and conduction of cardiac electrical activity. MicroRNA(miRNA),as an endogenous short-stranded non-coding RNA,negatively regulates gene expression post - transcriptionally and plays a key role in maintaining cardiac electrophysiological homeostasis and structural integrity. miRNA expression dysregulation is closely related to the pathogenesis of IA,and once miRNA function is disrupted,it directly impairs cardiac rhythm and contractile function,thereby inducing arrhythmia.Therefore,in - depth investigation of the expression patterns and regulatory mechanisms of cardiac - specific miRNAs is of great significance for revealing the pathological mechanisms of IA and developing novel therapeutic strategies.
