中文摘要: 骨质疏松型颈椎病(OCS)是一种以颈椎退行性变和骨代谢失衡为特征的复杂疾病,其发病机制涉及骨稳态破坏、椎间盘退变及神经血管压迫等多系统交互作用。Klotho是一种Ⅰ型跨膜蛋白,是成纤维细胞生长因子受体1(FGFR1)复合物的关键组成部分。近年来,Klotho蛋白因其在抗衰老、调控钙磷代谢及维持骨稳态中的核心作用成为研究焦点。最新研究证实,Klotho蛋白作为调节衰老和矿物质离子稳态的关键蛋白,可抑制炎症、抵抗肿瘤并调节内分泌系统,这些特性进一步突显了其作为临床骨修复理想分子的潜力。本文整合近5年研究进展,系统综述了Klotho通过成纤维细胞生长因子23(FGF23)信号轴在OCS中的作用,并探讨其作为治疗靶点的潜力,为OCS的精准干预提供了理论依据。
Abstract:Osteoporotic cervical spondylosis(OCS)is a complex disease characterized by cervical degenerative changes and bone metabolism imbalance, with its pathogenesis involving multi-system interactions such as disruption of bone homeostasis, intervertebral disc degeneration, and neurovascular compression. Klotho, a type Ⅰ transmembrane protein, serves as a critical component of the fibroblast growth factor receptor 1(FGFR1)complex. In recent years, Klotho protein has become a research focus due to its central roles in anti-aging effects, regulation of calcium-phosphorus metabolism, and maintenance of bone homeostasis. Emerging studies confirm that Klotho, a key regulator of aging and mineral ion homeostasis, exhibits properties including inflammation suppression, anti-tumor effects, and endocrine system modulation, further highlighting its potential as an ideal molecular candidate for clinical bone repair. This article synthesizes research advances from the past five years, systematically reviews Klotho??s role in OCS through the fibroblast growth factor 23(FGF23)signaling axis, and explores its therapeutic potential, thereby providing a theoretical foundation for precision intervention in OCS.
