中文摘要: 胶质母细胞瘤(GBM)是成年人最常见原发性脑肿瘤之一,发病后进展迅速且预后极差。GBM细胞中存在胆固醇代谢的重编程,涉及胆固醇的摄取、运输、酯化等过程。肿瘤细胞可利用胆固醇作为能量来源、构建细胞膜和介导信号分子等促进其增殖、进展和适应肿瘤微环境。近年研究表明GBM细胞中胆固醇代谢与患者预后紧密相关,其调节通路相关治疗可显著改善生存时间。本综述将回顾GBM中胆固醇代谢重编程现象及固醇调节元件结合蛋白、肝X受体、3-羟基-3-甲基戊二酰辅酶A还原酶(HMGCR)等在胆固醇代谢调节中的作用,并解读胆固醇代谢及调节通路相关靶向治疗的新进展。
Abstract:Glioblastoma(GBM)is one of the most common primary brain tumors in adults,characterized by rapid progression and extremely poor prognosis. Cholesterol metabolism reprogramming occurs in GBM cells,involving the processes of cholesterol uptake,transport and esterification. Tumor cells can use cholesterol as an energy source,construct cell membranes and mediate signal molecules to promote their proliferation,progression and adaptation to the tumor microenvironment. Recent studies have shown that cholesterol metabolism in GBM cells is closely related to patient prognosis,and therapies targeting its regulatory pathways can significantly improve survival time. This review summarizes the phenomenon of cholesterol metabolism reprogramming in GBM,the roles of sterol regulatory element-binding proteins,liver X receptors,3-hydroxy-3-methylglutaryl coenzyme A reductase(HMGCR)and others in the regulation of cholesterol metabolism,and interpret the latest advances in targeted therapies related to cholesterol metabolism and its regulatory pathways.
