中文摘要: 目的 探究结直肠癌(CRC)组织中植物同源结构域指蛋白(PHF1)、FBJ鼠科骨肉瘤病毒癌基因同源物B(FOSB)的表达与患者临床病理特征及预后的关系,为CRC的诊断及治疗提供参考。方法 选取2019年3月至2022年3月收集的河北北方学院附属第二医院124例CRC患者作为研究对象,收集患者术中的癌组织及癌旁组织,依据 3 年随访结果将患者分为存活组(95 例)及死亡组(29 例)。免疫组织化学法检测不同组织中PHF1、FOSB 表达水平。分析 PHF1、FOSB 表达与 CRC 患者临床病理特征的关系。分析不同预后 CRC 患者PHF1、FOSB 表达,采用多因素 Cox 回归分析 CRC 患者死亡的影响因素;Kaplan-Meier 法绘制生存曲线。结果与癌旁组织比较,癌组织中 PHF1、FOSB 阳性表达患者占比较高(P<0.05)。淋巴结转移及Ⅲ期的 CRC 患者PHF1、FOSB阳性表达率高于无淋巴结转移及Ⅰ~Ⅱ期的患者(P<0.05)。死亡组患者PHF1、FOSB阳性表达率高于存活组(P<0.05)。PHF1、FOSB表达是CRC患者死亡的独立危险因素(P<0.05)。PHF1阳性表达患者3年生存率低于阴性表达患者(71.59% vs 88.89%,log-rank χ2=4.711,P=0.030)。FOSB阳性表达患者3年生存率低于阴性表达(72.04% vs 90.32%,log-rank χ2=4.577,P=0.032)。结论 CRC组织中PHF1、FOSB均呈现高表达,与患者临床病理特征及预后联系密切。

Abstract:Objective To investigate the relationship between the expression of plant homeodomain finger protein 1(PHF1)and FBJ osteosarcoma viral oncogene homolog B(FOSB)in colorectal cancer(CRC)tissues and the clinical pathological features and prognosis of patients,so as to provide a reference for the diagnosis and treatment of CRC.Methods A total of 124 CRC patients collected in The Second Affiliated Hospital of Hebei North University from March 2019 to March 2022 were enrolled as study subjects. Intraoperative cancer tissue and adjacent tissue were collected from the patients. Based on the 3-year follow-up results,the patients were divided into a survival group(n=95)and a death group(n=29). Immunohistochemistry was used to detect the expression of PHF1 and FOSB in different tissues. The relationship between PHF1,FOSB expression and clinical pathological features of CRC patients was discussed. Expression levels of PHF1 and FOSB were compared between CRC patients with different prognoses.Multivariate Cox regression analysis was used to analyze the influencing factors of death in CRC patients. In addition,Kaplan-Meier method was used to plot survival curves. Results Compared with adjacent tissues,the proportions of positive PHF1 and FOSB in cancer tissues were higher(P<0.05). The proportions of positive PHF1 and FOSB in CRC patients with lymph node metastasis and stage Ⅲ were higher than those in patients with non-lymph node metastasis and stage Ⅰ-Ⅱ(P<0.05). The death group had higher positive rates of PHF1 and FOSB than the survival group(P<0.05).The PHF1 and FOSB were independent risk factors to the death of CRC patients(P<0.05). The positive PHF1 patients had a lower 3-year survival rate than negative PHF1 patients(71.59% vs 88.89%,log- rank χ2=4.711,P=0.030). In addition,the positive FOSB patients had a lower 3-year survival rate than negative FOSB patients(72.04% vs 90.32%,log- rank χ2=4.577,P=0.032). Conclusion PHF1 and FOSB are highly expressed in CRC tissues,which are closely related to the clinical pathological features and prognosis.
