中文摘要: 微卫星不稳定性(microsatellite instability,MSI)是指由于DNA错配修复系统功能缺陷导致微卫星区域长度或结构发生改变的现象,通常与肿瘤的发生、发展、预后和治疗密切相关。因此,MSI的检测对肿瘤治疗具有重要意义。目前,MSI的检测方法主要包括聚合酶链式反应法、免疫组织化学法和二代测序法。通过检测微卫星位点的变化或DNA错配修复系统蛋白的表达情况可以判断MSI的状态,从而指导肿瘤治疗。本文对MSI发生机制、检测方法及临床意义进行阐述。
Abstract:Microsatellite instability(MSI)refers to the phenomenon of the length or structure changes of microsatellite regions due to defects in the function of the DNA mismatch repair system,which is usually closely related to the development,progression,prognosis and treatment of tumors. Therefore,the detection of MSI is of great significance for tumor treatment. At present,MSI detection methods mainly include polymerase chain reaction,immunohistochemistry and next-generation sequencing. By detecting changes in microsatellite loci or the expression of DNA mismatch repair system proteins,the status of MSI can be determined,so as to guide tumor treatment. This article describes the mechanism and detection methods of MSI and the clinical significance of MSI detection.
