中文摘要: 目的 探讨子宫内膜异位症(EMT)中程序性坏死的作用,以及高迁移率族蛋白B1(HMGB1)与受体相互作用蛋白3(RIP3)之间的相互作用机制。 方法 2018年4月至2019年4月在徐州医科大学附属医院行子宫切除术的患者64例,将其中经妇科腹腔镜手术和术后病理诊断为EMT的患者43例分为观察组1(Ⅰ、Ⅱ期,22例)和观察组2(Ⅲ、Ⅳ期,21例);将非EMT患者经宫腔镜检查子宫内膜处于月经周期增殖晚期的5例作为正常组,处于月经周期分泌晚期的16例为坏死组。免疫荧光染色和Western blot检测4组对象子宫内膜组织中细胞程序性坏死相关蛋白RIP3和磷酸化混合系列激酶样结构域蛋白(p-MLKL)的表达水平;ELISA法检测血清中HMGB1的含量;Western blot检测HMGB1蛋白的表达水平;免疫共沉淀法验证RIP3蛋白和HMGB1蛋白的相互结合。 结果 观察组1、观察组2子宫内膜组织中p-MLKL和RIP3蛋白相对表达量较坏死组显著下降(P<0.05,P<0.01)。观察组1、观察组2和坏死组胞质中HMGB1水平较正常组显著增加(P<0.05,P<0.01)。观察组1、观察组2血清中HMGB1含量较正常组显著增加(P<0.05,P<0.01),但坏死组血清中HMGB1水平未显著升高。与正常组相比,坏死组中RIP3与HMGB1的结合水平显著升高,而观察组2中两者结合程度较低。 结论 EMT患者子宫内膜中细胞程序性坏死相关蛋白水平下降,且胞外HMGB1释放增多与RIP3-HMGB1的结合水平降低有关。RIP3与HMGB1的相互作用可能参与EMT的负向调控。
Abstract:Objective To investigate the role of necroptosis in endometriosis and the mechanism of interaction of high mobility group box-1 (HMGB1) protein and receptor interacting protein 3 (RIP3). Methods A total of 64 patients who underwent hysterectomy at the Affiliated Hospital of Xuzhou Medical University from April 2018 to April 2019 were enrolled.Forty-three patients with EMT diagnosed by gynecological laparoscopic surgery and postoperative pathology were divided into observation group 1 (stage Ⅰ,Ⅱ,22 cases) and observation group 2 (stage Ⅲ,Ⅳ,21 cases);5 cases of non-EMT endometrial proliferation in the late stage of menstrual cycle were taken as the normal group,and 16 in the late secretory stage of menstrual cycle were the necrosis group.Immunofluorescence staining and Western blot were used to detect the expression levels of programmed cell necrosis-related protein——RIP3 and phosphorylated mixed lineage kinase-domain like protein (p-MLKL) in endometrial tissue of the four groups.The serum HMGB1 content was detected by ELISA.Western blot was used to detect the expression levels of HMGB1;co-immunoprecipitation method was used to verify the mutual binding of RIP3 protein and HMGB1 protein. Results The relative expression level of p-MLKL and RIP3 protein in the endometrial tissues of observation group 1 and 2 were significantly lower than those in necrosis group (P<0.05,P<0.01).The level of HMGB1 in the cytoplasm of the observation group 1,2 and the necrotic group were significantly increased compared with the normal group (P<0.05,P<0.01).The level of HMGB1 in the serum of observation group 1 and 2 were significantly increased compared with the normal group (P<0.05,P<0.01),but the level of HMGB1 in the serum of the necrotic group did not increase significantly.Compared with the normal group,the binding level of RIP3 and HMGB1 was significantly increased in the necrotic group,while the binding degree of RIP3 and HMGB1 was lower in the observation group 2. Conclusions The level of programmed cell necrosis related proteins in the endometrium of EMT patients are decreased,and the increased release of extracellular HMGB1 is related to the decrease of the binding level of RIP3-HMGB1.The interaction between RIP3 and HMGB1 may be involved in the negative regulation of EMT.
