中文摘要: 肝细胞核因子4α(HNF4α)是肝细胞及肝细胞癌(HCC)代谢的核心转录因子,可调节多种肝脏代谢相关疾病的进展并导致HCC的发生。大量研究表明,HNF4α异常表达可导致癌细胞失控性增殖和代谢重编程。本文系统综述了HNF4α在糖代谢、脂质代谢、药物代谢、氨基酸代谢及凝血功能中的关键作用及其分子机制,总结HNF4α对参与各类代谢过程的肝脏特异性基因表达的多种调控机制,并探讨以其为靶点的潜在治疗策略,为代谢性疾病和HCC的临床干预提供新思路。
Abstract:Hepatocyte nuclear factor 4α(HNF4α)is a central transcription factor for the metabolism of hepatocytes and hepatocellular carcinoma(HCC). HNF4α can regulate the progression of diverse liver metabolism?related diseases and contribute to the development of HCC. Accumulating evidence demonstrates that aberrant expression of HNF4α drives uncontrolled cancer cell proliferation and metabolic reprogramming. This review systematically elucidates the pivotal roles and molecular mechanisms of HNF4α in glucose metabolism,lipid metabolism,drug metabolism,amino acid metabolism,and coagulation function. It also comprehensively summarizes the diverse regulatory mechanisms by which HNF4α modulates the expression of liver ?specific genes involved in various metabolic processes. Furthermore,this review explores potential therapeutic strategies targeting HNF4α,providing novel insights for clinical interventions in metabolic diseases and HCC.
