中文摘要: 目的 探讨食管鳞状细胞癌(ESCC)组织中甾醇 O-酰基转移酶 1(SOAT1)和髓样细胞触发受体 1(TREM1)mRNA表达水平,并分析其与ESCC患者临床特征和预后的关系。方法 纳入徐州医科大学附属医院于2017年1月至2019年6月首次就诊并接受初治的118例ESCC患者,收集患者术后切除的癌组织和癌旁正常组织样本,采用实时定量聚合酶链式反应(qRT-PCR)方法检测SOAT1和TREM1的mRNA表达水平;比较癌组织与癌旁组织SOAT1和TREM1的mRNA表达水平差异;比较不同临床特征患者癌组织中SOAT1和TREM1的mRNA表达水平差异;采用Cox回归分析影响ESCC患者预后的因素。结果 SOAT1与TREM1的mRNA在癌组织中的表达水平显著高于癌旁组织(P<0.05)。肿瘤最大直径>3 cm、病理为低分化与未分化、TNM分期Ⅲ期的ESCC患者癌组织SOAT1 mRNA相对表达水平显著增高(P<0.05)。有吸烟史、肿瘤最大直径>3 cm、病理为低分化与未分化、TNM分期Ⅲ期的ESCC患者癌组织TREM1 mRNA相对表达水平显著增高(P<0.05)。单因素与多因素Cox回归分析结果提示SOAT1 mRNA相对表达水平、TREM1 mRNA相对表达水平、肿瘤直径、分化程度以及TNM分期是影响ESCC患者预后的独立因素(P<0.05)。结论 ESCC组织中SOAT1与TREM1 mRNA表达水平显著上调,且SOAT1 mRNA相对表达水平、TREM1 mRNA相对表达水平是影响ESCC患者预后的独立危险因素。

Abstract:Objective To investigate the mRNA expression levels of sterol O-acyltransferase 1(SOAT1)and triggering receptor expressed on myeloid cells 1(TREM1)in esophageal squamous cell carcinoma(ESCC)tissues and analyze their associations with clinical characteristics and prognosis. Methods A total of 118 ESCC patients,who were first diagnosed and received initial treatment at the Affiliated Hospital of Xuzhou Medical University from January 2017 to June 2019,were included. Postoperative tumor tissues and adjacent normal tissues were collected. Quantitative real-time polymerase chain reaction (qRT-PCR) was used to detect SOAT1 and TREM1 mRNA expression levels.Differences in mRNA expression between tumor and adjacent tissues were compared. Associations between SOAT1 and TREM1 mRNA expression and clinical characteristics were analyzed. Cox regression was performed to identify prognostic factors. Results SOAT1 and TREM1 mRNA expression levels in tumor tissues were significantly higher than those in adjacent normal tissues(P<0.05). Higher SOAT1 mRNA expression was observed in patients with tumor diameter >3 cm,poorly/undifferentiated pathological differentiation,and TNM stage Ⅲ(P<0.05). Elevated TREM1 mRNA expression was associated with smoking history,tumor diameter >3 cm,poorly/undifferentiated pathology,and TNM stage Ⅲ(P<0.05). Univariate and multivariate Cox regression analyses indicated that SOAT1 mRNA expression,TREM1 mRNA expression,tumor diameter,differentiation grade,and TNM stage were independent prognostic factors.Conclusion SOAT1 and TREM1 mRNA expression levels are significantly upregulated in ESCC tissues,and both are independent risk factors for poor prognosis in ESCC patients.
